Melatonin downregulates TRPC6, impairing store-operated calcium entry in triple-negative breast cancer cells

چکیده

Melatonin has been reported to induce effective reduction in growth and development a variety of tumors, including breast cancer. In triple-negative cancer (TNBC) cells, melatonin attenuates features, such as tumor apoptosis resistance, through number still poorly characterized mechanisms. One biological process that is important for TNBC cells store-operated Ca2+ entry (SOCE), which modulated by TRPC6 expression function. We wondered whether might intersect with this pathway part its anticancer activity. show melatonin, the nanomolar range, significantly MDA-MB-231 cell viability, proliferation, migration time- concentration-dependent manner, without having any effect on nontumoral epithelial MCF10A cells. Pretreatment different concentrations reduced SOCE altering release from intracellular stores. By contrast, was unaffected melatonin. MDA-MB-468 line, not only attenuated migration, SOCE, but also or function channel Orai1. The exogenous overcame silencing inhibition impaired inhibitory SOCE. These findings indicate downregulation be involved melatonin's effects influx maintenance hallmarks point toward novel antitumoral mechanism

roles in regulating cell functions such as chemotaxis, motility or secretion. Stimulation by neurotransmitters or hormones activates intracellular production of inositol 1, 4, 5-trisphosphate (IP3) and diacylglycerol (DAG). IP3 binds to the IP3 receptor on the Ca store (endoplasmic reticulum ; ER) membrane and releases the sequestered Ca to the intracellular space. Increased intracellular Ca fu...

Phemindole, a Synthetic Di-indole Derivative Maneuvers the Store Operated Calcium Entry (SOCE) to Induce Potent Anti-Carcinogenic Activity in Human Triple Negative Breast Cancer Cells

Triple-negative breast cancer (TNBC), is a specific subtype of epithelial breast tumors that are immuno-histochemically negative for the protein expression of the estrogen receptor (ER), the progesterone receptor (PR) and lack over expression/gene amplification of HER2. This subtype of breast cancers is highly metastatic, shows poor prognosis and hence represents an important clinical challenge...

Requirement for store-operated calcium entry in sodium butyrate-induced apoptosis in human colon cancer cells.

The SOCE (store-operated Ca2+ entry) pathway plays a key role in both normal cells and cancerous cells. However, its molecular mechanism remains a long-lasting puzzle of Ca2+ signalling. In this paper, we provide evidence that butyric acid, a dietary fibre-derived short-chain fatty acid, induces apoptosis of colon cancer cells via SOCE signalling networks. We found that sodium butyrate (NaB) in...

Store-operated calcium entry and calcium influx via voltage-operated calcium channels regulate intracellular calcium oscillations in chondrogenic cells.

Chondrogenesis is known to be regulated by calcium-dependent signalling pathways in which temporal aspects of calcium homeostasis are of key importance. We aimed to better characterise calcium influx and release functions with respect to rapid calcium oscillations in cells of chondrifying chicken high density cultures. We found that differentiating chondrocytes express the α1 subunit of voltage...

STIM and Orai1 Variants in Store-Operated Calcium Entry

Store-operated Ca(2+) entry (SOCE) is an ubiquitous mechanism for Ca(2+) entry in eukaryotic cells. This route for Ca(2+) influx is regulated by the filling state of the intracellular Ca(2+) stores communicated to the plasma membrane channels by the proteins of the Stromal Interaction Molecule (STIM) family, STIM1, and STIM2. Store-dependent, STIM1-modulated, channels include the Ca(2+) release...
